What is chloroquine mode of action

Tell your doctor and pharmacist about all of your drugs (prescription or OTC, natural products, vitamins) and health problems. You must check to make sure that it is safe for you to take chloroquine with all of your drugs and health problems.

Chloroquine resistance is associated with a decrease in the amount of chloroquine that accumulates in the food vacuole, the site of action for chloroquine. The mechanism for this decreased accumulation is controversial. Some studies have shown that the decrease in drug accumulation is due to an increase in drug efflux. The drug chloroquine is bactericidal for Bacillus megaterium; it inhibits DNA and RNA biosynthesis and produces rapid degradation of ribosomes and dissimilation of ribosomal RNA. Inhibition of protein synthesis is also observed, evidently as a secondary effect. A severe eye problem has happened with chloroquine. This may lead to lasting eyesight problems. The risk may be higher if you have some types of eye or kidney problems. The risk may also be higher with some doses of chloroquine, if you use chloroquine for longer than 5 years, or if you take certain other drugs like tamoxifen.

For all uses of chloroquine: WARNING/CAUTION: Even though it may be rare, some people may have very bad and sometimes deadly side effects when taking a drug. Do not start, stop, or change the dose of any drug without checking with your doctor.

Chloroquine and hydroxychloroquine are drugs derived from the quinoline molecule. Both are used as antimalarial blood schizonticides, and hydroxychloroquine is also frequently used as an antirheumatic. Their mechanism of action is not entirely understood. Mechanism of Action Chloroquine, a 4-aminoquinoline, is an anti-protozoal agent. The precise mechanism by which Chloroquine exhibits activity is not known. Chloroquine, may exert its effect against Plasmodium species by concentrating in the acid vesicles of the parasite and by inhibiting polymerization of heme. It can also inhibit certain enzymes by its interaction with DNA. Chloroquine binds to heme or FP to form the FP-chloroquine complex; this complex is highly toxic to the cell and disrupts membrane function. Action of the toxic FP-chloroquine and FP results in cell lysis and ultimately parasite cell autodigestion. In essence, the parasite cell drowns in its own metabolic products.
