Toads fed chloroquine plus primaquine developed tumors in the liver, kidney, lung, and urinary bladder, and all the tumors were diagnosed as histiocytic sarcomas. It is speculated that one or more metabolites of chloroquine and primaquine eg, quinone may be responsible for tumor induction in the toads. In a previous nonimmune adult traveler study, chloroquine-proguanil recipients experienced more drug-related gastrointestinal events than did atovaquone-proguanil recipients and more frequently discontinued prophylaxis because of those events. In the present trial, the most frequently reported events in both cohorts were gastrointestinal complaints.
